Frequently Asked Questions

Q: How do I order DNA oligos?

For oligos not associated with a sequencing order, Log in to your account on www.etonbio.com. Select the “Place Order” option under the Oligo Synthesis heading and follow the steps through the order form.

If you would like to order primers to be used with your sequencing order, please select the “Place Order” option under the DNA Sequencing heading. On the Order Details page, check the box in the “Order Primer” column for reactions with primers that you need to have synthesized. This will bring up a window that will allow you to enter your primer name, sequence and choose a storage option. You only need to do this once for each primer you are ordering, even if it used multiple times within the same order.

Q: Can I use the same account information and PO for both sequencing and oligo synthesis?

Yes. If you would like to use a PO to purchase oligos, please enter the necessary information on the payment page of your order. If you are ordering oligos and sequencing together, then you have the option of entering a single PO for both the oligo synthesis and the sequencing or you can enter a different PO for each.

Q: Can I order oligos and sequencing together?

Yes, when placing your sequencing order check the box in the “Order Primer” column for reactions with primers that you need to have synthesized. Q: What is the cutoff time for placing an oligo order?

Oligos up to 25 nt can be ordered by 10 am for same day completion.

Oligos up to 60 nt can be placed before 6pm for next day completion. 

Any order placed after 6pm will be processed the next day.

Completion and delivery time will depend upon location and oligo length. See other FAQ questions for full details.

Q: When can I expect the sequencing reactions to be run with my synthesized oligo?

San Diego and Durham branch customers: Oligos ordered before 10:00am(local time) will be available the same day for sequencing use. Oligos ordered by 6pm(local time) will be available the following business day for sequencing use.

 

Newark and Boston branch customers: Oligos ordered by 10am PST will be available next business day for sequencing use.

 

All sequencing orders will be processed as soon as oligos are ready and samples are at our location.

Q: Will you store oligos I have synthesized for future sequencing reactions? For how long?

You have the option of storing synthesized primers for either two weeks or three months past the original order date. They can be used for additional reactions during this time window but all oligos will be discarded after three months.

Q: When can I expect my oligo to be delivered?

 Orders with large quantities, normalization and purification may take additional time for delivery. See below for information on our turnaround times based on lengths of your oligos. 

For oligos up to 25 nucleotides:

 

Sequencing Use:

San Diego branch customers: Oligos ordered before 10:00am(local time) will be available the same day for sequencing use. Oligos ordered by 8pm(local time) will be available the following business day for sequencing use.

Boston and Durham branch customers::  Oligos ordered before 10:00am(local time) will be available the same day for sequencing use. Oligos ordered by 6pm(local time) will be available the following business day for sequencing use.

Union branch customers:Oligos ordered before 11:00am(local time) will be available the same day for sequencing use. Oligos ordered by 8pm(local time) will be available the following business day for sequencing use.

Delivery:

San Diego and Union branch customers: Oligos ordered by 8pm(local time) will be delivered the next business day no later than 5pm.

Boston and Durham branch local customers: Oligos ordered by 6pm(local time) will be delivered the next business day no later than 5pm.

All non-local customers: Oligos ordered before 10:00am PST will be shipped out the same day for next business day delivery. Arrival times vary per delivery location. Orders placed after 10:00am PST will be shipped the following day with next business day delivery.

 

For oligos between 26 to 70 nucleotides:

San Diego and Union branch customers: Oligos ordered by 8pm(local time) will be delivered the next business day no later than 5pm.

Boston and Durham branch local customers::  Oligos ordered by 6pm(local time) will be delivered the next business day no later than 5pm.

All non-local customers: Oligos ordered before 6pm PST will be shipped out the next business day with over night delivery. Arrival times vary per delivery location.

 

For oligos 71 nucleotides and longer:

Turn around time will vary based on the length and scale of the oligo ordered. Please contact support@etonbio.com for an estimated delivery time.

 

For oligos that need purification:

Oligos needing HPLC and PAGE purification will be completed within 4-7 business days. For oligos longer than 70bp or large volume orders, turnaround time may be extended a few days. Oligo will be delivered within 1-2 business days after completion.

Q: In what form are oligos delivered?

 Oligos are delivered in their dry state.

Q: How should I store my oligo?

Oligonucleotides should be stored at -20 °C, preferentially in their dried form. Typically the molecule will be stable at this temperature for more than a year. Although oligonucleotides are stable in solution, they are also easily contaminated in this form, therefore Eton recommends that oligonucelotides being stored in solution are separated into aliquots. This not only helps to prevent contamination during repeated use but also protects the oligonucleotide from degradation due to repeated freezing and thawing. For your convenience, we provide four extra labels with the data sheet for your aliquot tubes.

Q: What length oligos do you offer?

We offer oligos between 15 and 150 nucleotides in length. For sequencing purposes, oligos should be between 18 and 24 nulceotides long with a GC content of 50%. If you would like more information about designing oligos to be used for sequencing please email support@etonbio.com.

Q: What synthesis scales do you offer?

We offer synthesis on 25nmol, 50nmol, 200nmol and 1umol scales. The synthesis scale refers to the amount of starting material used to synthesize the oligo not the amount of final product created. Because synthesis requires a series of steps and no chemical reaction is 100% efficient, we cannot predict the actual final yield, though we do guarantee a minimum final yield for each oligo.

Q: What oligo modifications do you offer?

Modifications can be incorporated into an oligo during the synthesis process. We currently offer are Phosphate and Biotin modifications on both the 5' and 3' ends of the oligo.

Q: What can modifications be used for?

Biotin is one of the most popular options. This modification can be used to link the oligo with another molecule. 

 

5' Phosphorylation can be added if an oligo will be used as a DNA ligase substrate.

 

3' Phosphorylation can be used to block extension by DNA polymerases. 

Q: How do I know how much DNA is actually present?

The microgram and nanomole amounts for each oligo you receive will be written on the label on the oligo tube as well as listed on the data sheet. The OD reading and molecular weight are also notated and can be used to calculate the amount of oligo present.

Q: How can I have my oligo delivered?

You must place your order for oligo synthesis using our Oligo ordering form.  All oligos ordered using our Oligo/Sequencing combination form will be stored in the lab for use.

Q: Will I be charged a shipping fee?

For all oligo orders pertaining to sequencing, no shipping fee applies. For oligo orders within branch drop off/delivery areas, no shipping fee applies. For oligo orders outside of our immediate delivery areas, a shipping fee will apply.

Q: Once my oligo has been shipped, when should I expect to receive it?

We ship all oligo orders to be delivered outside of our local delivery area overnight via FedEx or UPS.

Q: What concentration can you normalize to?

 We provide three normalized concentrations: 100 umol, 50umol and 25 umol.

Normalization may delay delivery of your oligo depending on the size of your order.

Q: What type of buffer are normalized oligos delivered in?

 You have a choice of TE buffer(default) or diH2O. If you need it for sequencing or PCR, diH2O is recommended.

Q: Do you charge for normalization?

 No. This service is free of charge.

Q: How do I request normalization on my oligo?

You will need to order your oligo through the oligo synthesis order form. Oligos ordered using our Sequencing/Oligo order form will not be given the option for normalization. Once in the oligo synthesis order form, you can select the concentration amount you would like your oligo to be normalized to.
